Undo, erase and clear
Undo with ⌘Z and redo with ⇧⌘Z while drawing, remove single strokes with the eraser, and wipe everything with ⌥⇧C.
While you’re drawing, ⌘Z undoes, ⇧⌘Z redoes and ⌥⇧C clears the screen. The eraser (0) removes individual strokes. The toolbar has Undo, Redo and Clear everything buttons too.

These shortcuts only belong to PenLayer while drawing is on. The moment you stop, ⌘Z goes back to your other apps. See Keyboard shortcuts for why, and how to change them.
Erasing and clearing can both be undone.
The eraser
Pick the eraser on the toolbar or press 0, then touch or drag over what you want gone. The eraser removes whole strokes, not pixels: touch any part of a stroke, shape, icon or badge and the whole thing goes.
How far the eraser reaches follows the current stroke weight.
On a graphics tablet, the pen’s erase button switches to the eraser for as long as you hold it. See Pen buttons.
Clear the screen
Press ⌥⇧C or use Clear everything on the toolbar to remove everything at once. If you clear by mistake, undo brings it back.
Undo after you stop drawing
By default, stopping throws away the drawing and its undo history. If you set When drawing stops to keep the drawing, undo still reaches back into the previous session when you start again. See Choose what happens when drawing stops.
